- APP (amyloid protein precursor) gene is on chromosome
21
- Pathological mutations in the Abeta coding sequence of
APP gene provoke familial Alzheimer's disease (see figure)
- Several regions of APP have trophic properties
- APP is a transmembrane protein, which is normally clived
by enzymes, named secretases.
- Secretases are responsible for the production of Abeta
peptides, of different lenghts (39 to 43 aminoacids)
- Secretases release sAPP, the extracellular part of APP,
which is likely to play important physiological roles, including trophic
activities
- Abeta 40 and 42 are the main components of amyloid plaques
- Abeta 42 is more directly linked to AD than Abeta 40
